چکیده

A new bioassay method for measurement of angiotensin II was clinically applied to determine the plasma renin activity (PRA) in normal subjects and hypertensive patients. Angiotensin II was extracted by a modified procedure reported by Boucher et al. Levels of angiotensin II extracted from 17 blood samples were independently determined by our new bioassay method and the rat pressor assay. The new Imethod could determine the minimum level of PRA of 0.25 ng. According to the new bioassay method, mean PRA levels for 9 normal subjects were 63.6±48.7 ngfdl, 177.7± 99.3 ngfdl, and 172.3±124.3 ngfdl at 1 hr after supine position, 1 hr after upright position with administration of 80 mg furosemide, and 2 hrs after upright position, respectively. Correlation coefficient of PRA levels between two bioassay methods was r=0.85 (p=0.005). It was concluded that the new method for PRA might be more reliable and much simpler than the rat pressor assay for clinical applications. PRA, Angiotensin II, Bioassay, Isolated cells.

Hyperreninemic hypoaldosteronism after chronic stress in the rat.

The effects of chronic stress on the renin-angiotensin-aldosterone system were studied by analysis of plasma hormone levels, kidney renin mRNA levels, adrenal angiotensin II receptors, and steroidogenesis in rats subjected to repeated immobilization (2 h daily) or intraperitoneal injections of 1.5 M NaCI for 14 d. 24 after the last stress in both stress models, plasma aldosterone levels were re...

Increased adrenal renin in transgenic hypertensive rats, TGR(mREN2)27, and its regulation by cAMP, angiotensin II, and calcium.

The newly established rat strain TGR(mREN2)27 is a monogenetic model in hypertension research. Microinjecting the mouse Ren-2d renin gene caused it to become a stable part of the genome. The rats are characterized by fulminant hypertension, low plasma active renin, suppressed kidney renin, high plasma inactive renin, and high extrarenal transgene expression, most prominently in the adrenal cort...

Estimation of plasma renin activity by radioimmunoassay of angiotensin I.

The renin-angiotensin-aldosterone hor monal system has been shown to be involved in the pathogenesis of certain hypertensive disorders, notably primary aldosteronism, malignant hypertension, renovascular hy pertension and that induced by oral con traceptives. 1 " 7 Plasma renin assay has be come an important diagnostic tool for in vestigating these disorders. Presence of a cellular renin angiotensin system in chromaffin cells of bovine adrenal medulla

The presence of a local renin-angiotensin system has been established in organs which serve as angiotensin targets. In this study, the expression of angiotensinogen mRNA, and subcellular localization of renin, angiotensin-converting enzyme and angiotensin II were investigated in bovine adrenal medullary cells in primary culture. By light microscopy, expression of angiotensinogen mRNA, immunorea...
